Multi-Modal Mass Spectrometric Imaging of Uveal Melanoma
نویسندگان
چکیده
Matrix assisted laser desorption ionisation mass spectrometry imaging (MALDI-MSI), was used to obtain images of lipids and metabolite distribution in formalin fixed embedded paraffin (FFPE) whole eye sections containing primary uveal melanomas (UM). Using this technique, it possible lysophosphatidylcholine (LPC) type lipid that highlighted the tumour regions. Laser ablation inductively coupled plasma (LA-ICP-MS) performed on UM showed increases copper within periphery intratumoural zinc tissue from patients with poor prognosis. These preliminary data indicate multi-modal MSI has potential provide insights into role trace metals cancer metastasis.

Uveal Melanoma
The eye’s uveal tract consists of the iris, ciliary body, and choroid. It contains a population of melanocytes. Uveal melanomas develop from melanocytes that reside within the stroma of the choroid, ciliary body or iris. The choroid is the vascular part of the human eye between retina and sclera. Though accurate measurements of the choroidal blood flow is difficult, it is known that choroid is ...

BACKGROUND: Malignant melanoma of the uveal tract is a rare malignancy but a significant cause of mortality and visual loss. Approximately 50% of patients diagnosed with a melanoma of the choroid or ciliary body will die of the disease within 15 years of enucleation. METHODS: The author reviewed the current literature on the clinical findings, epidemiology, and treatment of uveal melanoma. RESU...

Uveal melanoma is the most common primary intraocular malignancy and the leading primary intraocular disease which can be fatal in adults. In this paper epidemiologic, pathogenetic, and clinical aspects of uveal melanoma are discussed. Despite the advance in local ocular treatments, there has been no change in patient survival for three decades. Development of metastases affects prognosis signi...

Secondary ion mass spectrometry (SIMS) is capable of imaging biological molecules across single cells with several hundred nanometer lateral resolution, and has the potential to examine the functional segregation of lipids in cell membranes. Lipids are believed to play a crucial role in fundamental biological functions, such as intercellular communication. For example, high curvature lipids may...
متن کاملGenetics of Uveal Melanoma
Uveal melanoma is the most common type of primary eye cancer in adults, affecting 0.7/100,000 of the Western population yearly (Egan et al., 1988). The melanoma originates from neural crest derived melanocytes of the uvea (choroid, ciliary body and iris) and despite enucleation or conservative treatment half of patients die of, most often late appearing, metastatic disease (15-year survival: 53...
